My husband has been relocated with his job due to a company surplus. I had to resign to relocate our family. Can I file for unemployment in my new City? This is an in state move

Moving to relocate with a spouse is not good cause for quitting in Florida, but you should also check the FL statutes for yourself to confirm.
There is absolutely nothing restricting you from filing a claim .. just that I believe it will result in benefits being denied for a quit without good cause .. because it is not attributable to the employer and Florida did not have a provision creating an exception for this reason last time I looked.
